M13K07 genome
M13 is a filamentous phage that infects E. coli. It packages a single stranded genome of ~6400 bases, encoding 11 ORFs. The M13K07 sequence entered here includes the entire M13 genome as well as a sequence from pACYC177 bearing the p15a bacterial origin of replication and a kanamycin resistance gene. These additional sequences were inserted into the packaging sequence of M13, enabling the phage to propagate as a Kan-selectable plasmid. The modification does attenuate phage production though plaques are still detectable on NEB strain ER2267 (genotype: : F´ proA+B+ lacIq Δ(lacZ)M15 zzf::mini-Tn10 (KanR)/ Δ(argF-lacZ)U169 glnV44 e14-(McrA-) rfbD1? recA1 relA1? endA1 spoT1? thi-1 Δ(mcrC-mrr)114::IS10). In the presence of a second plasmid bearing the M13 packaging sequence and origin, M13K07 serves as a "helper phage," providing the proteins for the phage coat but allowing the alternative plasmid to be preferentially placed in the phage capsid.
