BioBrick base vector, as originally designed
BBa_I51000 is the original design of the BioBrick base vector BBa_I51020.
Synthesis of BBa_I51000 was problematic. We encountered two types of issues while trying to construct BBa_I51000. Faulty part design adversely impacted the synthesis process and problems during synthesis ended up compromising the design of our parts. Our synthesis difficulties largely stemmed from the dependence of commercial DNA synthesis processes on cloning, assembly and propagation of synthesized DNA in E. coli.
The BioBrick base vector that was successfully constructed is BBa_I51020.
